摘要:

Whole-Tissue Volume Imaging to Decipher Glial Response to Traumatic Neural Injury Microglia are the specialized immune cells of the central nervous system, and they play critical roles in various neuropathological conditions. However, the regulatory mechanism of microglial activation remains to be better understood. Here, we report the 3D volume fluorescence imaging of neurodegeneration-induced microglial activation by exploiting the optimized iDISCO technique. We observe that transcription factors IRF8 and PU.1 are highly up-regulated in microglial activation. By profiling their transcriptional landscapes, the cross-regulation mechanism between IRF8 and PU.1 is revealed. In addition, IRF8 and PU.1 act cooperatively through the composite IRF-ETS motifs that are specifically enriched on microglial activation-related genes. The formation of a ternary complex comprising the composite-motif DNA, IRF8 and PU.1 is biochemically verified. Furthermore, the specific deletion of IRF8 or PU.1 in microglia effectively abolishes their activation process. We have therefore demonstrated the central role of the transcriptional module of IRF8 and PU.1 in microglial activation under neuropathological conditions.

杨竞,北京大学生命科学学院Mcgovern脑科学研究所助理教授。北京大学本科毕业之后去美国德克萨斯西南医学中心攻读博士学位。先后在Genentech IncRockefeller 大学从事长达六年的博士后研究。他关于神经退行性疾病分子机制的研究获得国家自然科学基金的重点资助。他的研究方向主要是对TGFbeta在神经退行性疾病在小胶质细胞活化中的功能研究。迄今为止,杨竞博士在顶尖期刊上发表了多篇杰出的著作,包括CellScienceProc Natl Acad Sci USAAm J Physiol Endocrinol MetabJ Neurosci等。
