报告题目:Construction and Validation of a Real-Time In Vivo Biomolecular Interaction Imaging Platform Based on Excited-State Kinetics

Analyzing protein interactions in their native physiological context remains challenging, as current methods overwhelmingly rely on invasive ex vivo assays. Here, we present real-time transient absorption spectral imaging (TASI) as a label-free platform to resolve biomolecular interactions in vivo by exploiting their distinct excited-state kinetic signatures. By integrating a high-speed angle-to-delay converter, TASI achieves ultrafast acquisition (62.5 µs per transient absorption spectrum) alongside cellular morphological mapping. We validated this platform by quantifying in vivo glycated hemoglobin (HbA1c) within circulating red blood cells. TASI robustly differentiates the relaxation dynamics of glycated and native hemoglobin arising from glucose-induced conformational perturbations. In diabetic rats, TASI quantified in vivo HbA1c across the 4-15% clinical range, exhibiting high concordance with boronate affinity chromatography (Pearson's r = 0.982; mean relative error = 6.18%) and distinguishing hyperglycemic rats with 87% accuracy. TASI establishes a general framework for studying protein-molecule interactions in vivo, with potential applications in disease diagnosis and longitudinal monitoring.

个人简介 :


Dr. Wang Pu is currently a Professor at the School of Biological Science and Medical Engineering, Beihang University, a Research Fellow at the Beijing Advanced Innovation Center for Biomedical Engineering, a doctoral supervisor, and CEO of Zhendian Technology.
He earned his bachelor's degree from the Department of Physics, Fudan University. From 2009 to 2014, he pursued his doctoral studies at the Weldon School of Biomedical Engineering, Purdue University, under the supervision of Prof. Cheng Jixin, a leading expert in nonlinear imaging. During his PhD research, his main focus lay on the development of biophotonic medical devices, as well as the design and application of nonlinear microscopes.
Dr. Wang has published over 20 SCI papers and obtained 5 patents. As the first author or corresponding author, he has published research findings in top-tier journals including Nature Photonics, Science Advances, Light: Science & Applications and Nano Letters. He has led multiple Small Business Innovation Research/Small Business Technology Transfer (SBIR/STTR) programs in the United States and guided his team to accomplish a number of research and industrial transformation projects. Representative achievements include the industrialization of coherent Raman microscopes, and the translational development of photoacoustic imaging devices for breast and cardiovascular applications.
At present, his primary research directions cover the development of nonlinear Raman microscopes and their characterization applications for advanced materials and single-cell metabolism, as well as the biomedical applications of photoacoustic devices.
